Effect of Ticagrelor Variable Doses on the Pharmacokinetic Parameters of Tacrolimus in Rabbits |

Tacrolimus (TAC) is a potent immunosuppressant drug which is used as a prophylaxis treatment mainly in kidney transplantation to prevent organ rejection. TAC is predominantly metabolized by CYP3A4 and it is also likely to be a substrate for the P-glycoprotein (P-gp) efflux pump. Ticagrelor (TICA) is a new modified antiplatelet drug that has been shown to be a weak inhibitor for both CYP3A4 isoenzyme and P-gp. The purpose of this study is to evaluate the effect of TICA at various doses on TAC pharmacokinetic (PK) parameters in rabbits. An in vivo parallel-randomized drug-drug interaction study was conducted using fifteen healthy male rabbits divided into three groups (n=5 rabbits for each). In the control group, TAC (0.6 mg/kg/day) was administered as a single daily oral dose for six consecutive days, while in the test groups, TAC (0.6 mg/kg/day) was concomitantly co-administered with TICA at concentrations of 10 or 20 mg/kg/day. |
